Quantitative Analysis of Goose and Duck Down Blends with Real-time PCR

Abstract: In this project, a quantitative method based on real-time PCR technique was developed to determine the weight of goose down and duck down in down products. Two sets of primers and TaqMan probes that can react specifically to goose and duck mitochondrial DNA were designed, and they exert almost the same amplification rate. The two systems were used to establish the quantitative standard curve of down species determination(R2>0.99). Goose and duck down blends collected from different producing areas were employed to verify the method, and satisfactory results were obtained, which proved that the copy number of mitochondrion has little impact on quantifying results. The method can be applied to quick and large-batch quantitative identification of down species in down products.
